Description of Human BMP-7 / OP-1 (glycosylated, CHO)

Recombinant Human BMP-7 is 139 amino acid peptide which has due to glycosylation a molecular mass of 25-40kDa. It is produced ion CHO cells by expression of N-TERMINAL---Human BMP-2 (Met 1 – Arg 282) Human BMP-7 (Ser 293 – Arg 431)---C-TERMINAL. The DNA sequence encoding the human BMP-2 signal peptide and propeptide (1"282 amino acid) fused to the human rhBMP-7 mature chain (293"431 amino acid). The mature recombinant BMP-7 is generated by proteolytic removal of the signal and the propeptide. TGF-ß family members are key modulators of cell proliferation, differentiation, matrix synthesis, and apoptosis. As implied by their name, BMPs initiate, promote, and regulate the development, growth, and remodeling of bone and cartilage. In addition to this role, BMPs are also involved in prenatal development and postnatal growth, remodeling, and maintenance of a variety of other tissues and organs. BMP-7, also known as osteogenic protein-1 or OP-1, is a potent bone inducing agent, which in the presence of an appropriate osteoconductive carrier (e.g. collagen sponge or synthetic hydroxyapatite) can be used in the treatment of bone defects. A bone-graft substitute, called OP-1TM implant, made of recombinant human BMP-7 associated with bovine bone-derived collagen, has recently been approved by the FDA as a device for treating critical-size bone fractures. The potential use of BMP-7, especially the monomer from E.Coli, in dental reconstructive surgeries is currently under investigation.

Biological Activity

The ED₅₀ of ≤ 100ng/ml was determined by an alkaline Phosphatase Assay using MC3T3-E1 cells. It corresponds to a specific activity of ≥ 1x10⁴ units/mg. If ATDC-% cells are used the ED₅₀ is 0.02-0.04.

Reconstitution

We recommend a quick spin followed by reconstitution in water to a concentration of 0.5 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week or in working aliquots at -20°C to -80°C. Working aliquots should be at the highest practical concentration. For extended storage, it is recommended to further dilute in a buffer containing a carrier (e.g. 0.1% HSA order number: 2835.955.xyz or 2835.958.xyz) and store in working aliquots at -20°C to -80°C.

Stability

The lyophilized protein is stable at room temperature for up 1 month and at least until the lot specific expiry date if kept below -18°C. Reconstituted sTRAIL is stable for at least 6 months when stored in working aliquots at -20°C to -80°C. Please avoid repeated freeze-thaw cycles.
