Description of Murine GM-CSF
Recombinant murine Granulocyte Macrophage Colony Stimulating factor (mGM-CSF), derived from E.coli, is a 14.2 kDa protein consisting of 125 amino acids. Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a monomeric glycoprotein cytokine that regulates the production, differentiation, and function of macrophages and granulocytes. The gene encoding GM-CSF is located in the chromosome region 5q31, within a cluster of related genes including interleukins 4, 5, and 13. Deletions in this region have been associated with the 5q- syndrome and acute myeloid leukemia.GM-CSF plays a central role in maintaining lung homeostasis. Mice deficient in GM-CSF develop pulmonary alveolar proteinosis due to impaired surfactant clearance. In the context of SARS-CoV-2 infections, studies have shown elevated GM-CSF levels in patients with acute respiratory distress syndrome (ARDS), contributing to cytokine storm-mediated hyperinflammation. GM-CSF activates M1-type macrophages and enhances the release of pro-inflammatory cytokines such as IL-1β, IL-6, and TNF-α. Despite its pathological role, GM-CSF is physiologically crucial for the differentiation of alveolar macrophages, viral clearance, and epithelial regeneration. However, clinical trials with GM-CSF blockers like Gimsilumab have not shown significant improvement in COVID-19 survival rates, indicating complex biphasic mechanisms. Therapeutically, early GM-CSF administration could enhance immune responses, while late blockade might dampen hyperinflammation. GM-CSF also exhibits immunomodulatory duality: it exacerbates autoimmune processes in rheumatoid arthritis but can promote regulatory T-cell responses in other contexts. This Janus-faced function underscores the need for context-dependent therapeutic manipulation.

Biological Activity
The ED50 of ≤ 0.2 ng/ml was determined by the dose-dependent stimulation of the proliferation of murine FDC-P1 cells. It corresponds to a specific activity of ≥ 5x106 units/mg.
Cross Reactivity
Mouse GM-CSF has been reported to be partially cross-reactive with rat GM-CSF, making it functional in some experimental contexts involving rat cells, though species-specific cytokines remain ideal for precise applications.
Reconstitution
We recommend a quick spin followed by reconstitution in sterile 20mM AcOH (acetic acid) to a concentration of at least 100µg/ml. This solution can then be diluted into other aqueous solutions and stored at 4°C for 1 week or at -20°C for future use.
Stability
The lyophilized protein is stable at room temperature for a few weeks, but best stored at -20°C. Reconstituted GM CSF should be stored in working aliquots at -20°C. We recommend the addition of a carrier (1% HSA e.g. 2835.958.xyz, BSA) for long term storage of the reconstituted protein. Please avoid repeated freeze-thaw cycles.
