Description of Human DPP4 / CD26 (recombinant)

Recombinant human DPP4 (rhDPP4) produced in Insect cells is a single, glycosylated protein consoting of 737 amino acids (aa 39-766) with a molecular mass of 85.4 kDa and fused to a 6  aa His Tag at C-terminus. DPP4 (dipeptidyl peptidase IV, DPP IV, CD26), also known as adenosine deaminase (ADA)-binding protein and T-cell activation antigen CD26, is a homodimeric atypical serine protease belonging to the prolyl oligopeptidase family. It is expressed on T lymphocytes, where its expression is upregulated upon activation, as well as on activated B cells, natural killer cells, epithelial cells, and fibroblast-related cell types. As an intrinsic membrane glycoprotein, CD26 functions as a serine exopeptidase that specifically cleaves X-proline or X-Alanin dipeptides from the N-terminus of polypeptides. Beyond its enzymatic activity, CD26 is involved in a wide range of biological processes, including T-cell activation, immune regulation, cell adhesion through interactions with extracellular matrix proteins such as fibronectin and collagen, and has been implicated in HIV infection. Altered CD26 peptidase activity has been reported in numerous tumor entities and correlates with tumor grade and disease severity in a subset of cancers, with CD26 acting either as a marker of malignancy or as a tumor suppressor depending on tumor type. Furthermore, changes in CD26 expression have been reported on circulating monocytes and CD4+ T cells in autoimmune diseases, including rheumatoid arthritis and systemic lupus erythematosus, highlighting its importance as a biomarker and potential therapeutic target in cancer, immunology, and inflammatory disease research.

 

Biological Activity

Biological activity is > 200 units/mg, where one unit is defined as the hydrolyzation of 1 µmole of p-nitroaniline per minute at pH 8.0 and 37°C, using 1 mM Gly-Pro p-nitroanilide as substrate.

 

Stability

Store at 4°C if entire vial will be used within 2-4 weeks. Store, frozen at -20°C for longer periods of time. For long term storage it is recommended to add a carrier protein (0.1% HSA, order number: 2835.955.xyz or 2835.958.xyz, or BSA, order number: 2835.919.xyz). Please avoid repeated freeze-thaw cycles.
