Description of Human Plasmin (Plasma)

Human Plasmin from human plasma is an enzyme with a serine type endopeptidase activity and a molecular mass of 81 kDa. It has been prepared from streptokinase activated human plasma Plasminogen. Plasmin plays an important role in the fibrinolytic cascade, in inflammation processes and wound healing. It degrades fibrin, the main component of blood clots. Generally, Plasmin catalysis the hydrolysis of peptides, arginine and lysine esters and converts fibrin into soluble products. For the preferential cleavage at the carboxyl side of lysin and arginine it shows a higher selectivity than trypsin. Plasmin has been found to activate the lymphangiogenic growth factors VEGF-C and VEGF-D (Bradley et. al, PMID: 12963694), while it degrades VEGF165 (Künnapuu et.al PMID: 33672235).

 

Human Virus Test

FDA approved Plasma from each donor has been tested and found negative for antibodies to HIV-1, HIV-2, HCV, HBc, and non-reactive for HBsAg.

 

Biological Activity

The biological active is ≥ 10 units/mg. One unit is defined as the amount of enzyme that hydrolyzes one µmole of tosyl-Gly-Pro-Lys-pNA per minute at 25°C, pH 7.8. One Unit = 1.25 CU.

 

Reconstitution

We recommend a quick spin followed by addition of x µl water (de-ionized) to obtain a solution with xmg/ml. Mix gently to assure adequate homogenization. Do not vortex. This solution can be stored at 2-8°C for up to 1 week or in working aliquots at -20°C. For further dilution please use the lyophilization buffer. Working aliquots should be at the highest practical concentration.

 

Stability

The lyophilized protein is stable at room temperature for up to 1 month and at least until the lot specific expiry date if kept below -18°C. Reconstituted Plasmin should be stored in working aliquots at -20°C. Please avoid repeated freeze-thaw cycles.
