Description of Human FGF-acidic / FGF-1
Recombinant human Fibroblast Growth Factor-acidic (hFGF-acidic) is a 15.8 kDa protein consisting of 140 amino acid residues, 141 including the Mat the start. FGF-acidic is one of 23 known members of the FGF family. Proteins of this family play a central role during prenatal development and postnatal growth and regeneration of a variety of tissues, by promoting cellular proliferation and differentiation. FGF-acidic is a non-glycosylated heparin binding growth factor that is expressed in the brain, kidney, retina, smooth muscle cells, bone matrix, osteoblasts, astrocytes and endothelial cells. FGF-acidic has the ability to signal through all the FGF-receptors.
Biological Activity
The ED50 of ≤ 0.5ng/ml was determined by the dose-dependent proliferation of mouse BALB/c 3T3 cells. It corresponds to a specific activity of ≥ 2 x 106 units/mg.
Reconstitution
We recommend a quick spin – do not vortex – followed by reconstitution in sterile water at 4°C to a concentration of 0.1 - 1.0mg/ml. Allow sample to sit for 5 min. at 4°C, spin to remove precipitant. This solution can then be diluted into other aqueous buffers and stored at 4°C for 1 week or -20°C for future use.
Stability
The lyophilized protein is stable for a few weeks at room temperature, but best stored at -20°C. Reconstituted FGF-acidic should be stored in working aliquots at -20°C. Please avoid repeated freeze-thaw cycles.
