Description of Human GDF-15 / MIC-1
Recombinant human GDF-15/MIC-1 is a disulfide linked homodimeric protein consisting of two 113 amino-acid poly-peptide chains, having a molecular weight of 24.8 kDa. GDF-15 belongs to the TGF-ß cytokine family, whose mem-bers play an important role during prenatal development and postnatal growth, and the remodelling and maintenance of a variety of tissues and organs. GDF-15 is expressed predominantly in the placenta and, to a much lesser extent, in various other tissues. The presence of GDF-15 in amniotic fluid and its elevated levels in the sera of pregnant women suggest GDF-15s involvement in gestation and embryonic development. GDF-15 generally exerts tumor suppressive activities and is one of the predominant factors produced and secreted in response to activation of the p53 pathway. Interestingly, the serum level of GDF-15 is positively correlated with neoplastic progression of several tumor types, including certain colorectal, pancreatic and prostate cancers.
Biological Activity
An ED50 of 1-2µg/ml was determined by its ability to inhibit DU-145 cells. An ED50 of 0,2-2 ng/ml was measured by its ability to activate SRE-SEAP reporter in HEK293 human embryonic kidney cells transfected with human c-Ret and human GFRAL.
Reconstitution
We recommend a quick spin followed by reconstitution in sterile filtered water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, we recommend to fur-ther dilute into a buffer containing a carrier protein (e.g. 0.1% HSA, order number: 2835.955.xyz or 2835.958.xyz, or BSA, order number: 2835.919.xyz) and store in working aliquots at -20°C to -80°C.
Stability
Lyophilized human GDF-15, although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution GDF-15 can be stored at 4°C for 2-7 days and for future use below -18°C. Please avoid repeated freeze-thaw cycles.
