Description of Human IFN-lambda-1 / IL-29
Recombinant Human IFN-lambda-1/IL-29 derived from E. coli is a 20 kDa single, non-glycosylated, protein consist-ing of 181 amino acids. It is distantly related to type I Interferons (IFNs) and the IL-10 family. Expression of Human IFN-lambda-1 (Interleukin 29) is induced by viral infection which interacts with a heterodimeric class II cytokine re-ceptor that consists of interleukin 10 receptor-beta (IL10rb) and interleukin 28 receptor-alpha. Human IFN-lambda-1 (Interleukin 29) exhibits common features with type I IFNs such as antiviral activity, antiproliferative activity and in vivo antitumor activity. Even so it acts similarly to IFNs, it is generally less effective and has activity only in a more limited range of cell lines. The genes for IFN-lambda-1, IFN-lambda-2 and IFN-lambda-3 are closely positioned on human chromosome 19. Human IFN-lambda-1 (Interleukin 29) induces ELR(-) CXC-chemokine mRNA in human peripheral blood mononuclear cells, in an IFN-gamma-independent manner. It can generate tolerogenic Dendritic Cells (DCs), an activity that could thwart IFN-beta functions. If Human IFN-lambda-1 (Interleukin 29) is produced in response to viral infection, it activates both monocytes and macrophages producing a restricted panel of cytokines and therefore is an important factor in activating innate immune responses at the site of viral infection. IFN-Lambda 1 antiviral and antiproliferative activity requires IFN-Lambda 2 receptor tyrosine residues.
Biological Activity
An ED50 of 0.2-0.5 ng/ml was found by testing its ability to activate STAT phosphorylation in an ISRE Luciferase Reporter Assay using human colon carcinoma COLO205 cells.
Reconstitution
We recommend a quick spin followed by reconstitution in sterile water to a concentration of at least 100 µg/ml, which can then be further diluted to other aqueous solutions. Do not vortex.
Stability
Lyophilized IFN-Lambda-1 is stable at room temperature for up to 3 weeks, but best stored below -18°C. Reconstitut-ed IFN-Lambda-1 can be stored at 4°C for 2-7 days and for future use below -18°C. For long term storage it is rec-ommended to add a carrier protein (0.1% HSA, order number: 2835.955.xyz or 2835.958.xyz, or BSA, order number: 2835.919.xyz). Please avoid repeated freeze-thaw cycles.
