Description of Human Immunoglobulin A1 (IgA1)
Human Immunoglobulin A1 (IgA1) is a 160 kDa protein. Serum IgA1 is the predominant subclass of Immunoglobulin A in human circulation, comprising approximately 90% of total serum IgA. It eatures a distinctive hinge region with multiple O-linked glycans, distinguishing it structurally from IgA2. IgA1 serves critical immune functions by neutralizing pathogens, facilitating immune complex clearance, and mediating antibody-dependent cell-mediated cytotoxicity through FcαRI receptors on neutrophils and monocytes. Aberrant glycosylation of IgA1 plays a central role in IgA nephropathy pathogenesis, where galactose-deficient IgA1 forms immune complexes that deposit in glomeruli, triggering kidney inflammation and damage. Additionally, IgA1 abnormalities are implicated in IgA vasculitis, celiac disease, and certain autoimmune conditions. Clinically, serum IgA1 measurements assist in diagnosing IgA-related disorders, monitoring disease progression, and evaluating treatment efficacy. Recent therapeutic approaches target IgA1 pathways in nephropathy, including proteasome inhibitors and complement pathway modulators. While IgA1 predominates in serum, secretory IgA shows a more balanced distribution between IgA1 and IgA2 (approximately 50:50), reflecting their specialized mucosal defense functions.
