Description of Human Lipoprotein a, [Lp(a)]

Human Lipoprotein a, [Lp(a)] is a 280000-700000 Da protein. Lipoprotein(a), or Lp(a), is a specialized lipoprotein particle found in plasma at concentrations typically ranging from 20 to 760 mg/L, with levels largely determined by genetic factors1. Structurally, Lp(a) consists of an LDL-like core bound to apolipoprotein(a), which imparts unique proatherogenic and prothrombotic properties. Elevated Lp(a) is an independent and causal risk factor for atherosclerotic cardiovascular diseases, including coronary artery disease, heart attack, stroke, aortic stenosis, and peripheral artery disease. Lp(a) promotes vascular inflammation and plaque formation by accumulating in vessel walls and carrying oxidized phospholipids, which trigger inflammatory responses and calcification67. It also interferes with fibrinolysis, increasing the risk of thrombosis. Currently, no FDA-approved therapies specifically target Lp(a) reduction, but its measurement can help identify individuals at high cardiovascular risk who may benefit from intensified preventive strategies. Research into targeted Lp(a)-lowering treatments is ongoing, offering hope for future clinical applications.
