Description of Human Urokinase

Human Urokinase (native) derived from human urine is a 54 kDa two-chain glycoprotein containing 411 amino acids with 12 disulfide bonds. Urokinase (UK) is a serine protease, which is one of the biological plasminogen activators. It is involved in several biological functions including fibrinolysis, embryogenesis, cell migration, tissue remodeling, ovulation, and wound healing. Urokinase is inhibited by the plasminogen activator inhibitors PAI-1 (1797.950.xyz) and PAI-2 (1798.950.xyz). It has been found that the expression of Urokinase and PAI-1 is increased in some cancer patients.

Human Virus Test

Urine has been tested and found negative for antibodies to HIV-1, HIV-2, HCV, and non-reactive for HBsAg.

Biological Activity

The biological activity is ≥ 150 000 IU/mg. One unit is defined as the amount of enzyme equal to an international standard tested by the fibrinolytic method of Johnson, et al. PMID: 5791803.

Reconstitution

We recommend a quick spin followed by reconstitution in sterile water to a concentration of at least 100µg/ml, which can then be further diluted to other aqueous solutions. Do not vortex.

Stability

Lyophilized Urokinase is stable at room temperature for up to 3 weeks, and at least until the lot specific expiry date if stored below -18°C. Upon reconstitution Urokinase can be stored at 4°C for 2-7 days and below -18°C for future use. Please avoid repeated freeze-thaw cycles.
