Description of Human BD-2

Recombinant Human Beta defensin-2 (BD-2) is a 41 amino acid peptide, with a molecular mass of 4,3 kDa. All Defensins are highly similar in their protein sequence. They are microbicidal and cytotoxic peptides made by neutrophils. The alpha-defensins are distinguished from the beta-defensins by the pairing of their three disulfide bonds. To date, six human beta-defensins have been identified: BD-1, BD-2, BD-3, BD-4, BD-5 and BD-6. Human BD-1 and Human BD-2 are monomeric, while Human BD-3 is a dimer. Defensins can act as chemoattractants towards immature dendritic cells and memory T-cells. The beta-defensin proteins are expressed as the C-terminal portion of precursors, they are released by proteolytic cleavage of a signal sequence and, in some cases, a propeptide sequence.

Biological Activity

The ED₅₀ of 10-100ng/ml was determined by the ability to chemoattract immature human dendritic cells.

Reconstitution

We recommend a quick spin followed by reconstitution in water to a concentration of at least 100 µg/ml, which can then be further diluted. Do not vortex. This solution can be stored at 2-8°C for up to 1 week or in working aliquots at -20°C to -80°C. Working aliquots should be at the highest practical concentration.

Stability

The lyophilized protein is stable at room temperature for up to 1 month and at least until the lot specific expiry date if kept below -18°C. Reconstituted BD-2 should be stored in working aliquots at -20°C to -80°C. Please avoid repeated freeze-thaw cycles.
