Description of Human CXCL16

Recombinant human CXCL16 is a 10.1 kDa protein containing 89 amino acids. It is a member of the CXC chemo-kine family and signals through the CXCR6 receptor. CXCL16 may play a role in attracting lymphocyte subsets dur-ing inflammation and may facilitate certain immune responses. The chemokine domain of CXCL16 contains six cysteine residues including the four highly conserved cysteine residues characteristic of CXC chemokines. The CXCL16 gene codes for a 273 amino acid polypeptide, which includes a 29 amino acid cytoplasmic domain and transmembrane sequence containing approximately 20 amino acids. The extracellular portion of CXCL16 contains a chemokines domain and an extended C-terminal “mucin-like stalk” sequence. The extracellular domain contains 89 amino acid residues (86 a.a. for the murine homolog).

Biological Activity

The ED₅₀ of ≤ 12ng/ml was measured by its ability to chemoattract mouse CXCR6-transfected mouse BaF3 cells.

Reconstitution

We recommend a quick spin followed by reconstitution in water to a concentration of 0.1-1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at 4°C for 1 week or -20°C for future use.

Stability

Lyophilized human CXCL16, although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution CXCL16 can be stored at 4°C between 2-7 days and for future use below -18°C.For long term storage we recommend to add a carrier protein (0.1% HSA, order number: 2835.955.xyz or 2835.958.xyz, or BSA, order number: 2835.919.xyz). Please avoid repeated freeze-thaw cycles.
