Description of Human Flt3-Ligand

Recombinant human Flt3-ligand is a soluble 17.6 kDa polypeptide containing 155 amino acid residues. Flt3-ligand is a growth factor proliferation of early hematopoietic cells. It binds to cells expressing the tyrosine kinase receptor Flt3. Flt3-ligand, by itself does not stimulate proliferation of early hematopoietic cells, but synergizes with other CSFs and interleukins to induce growth and differentiation. Unlike SCF, Flt3-ligand exerts no activity on mast cells. Multiple isoforms of Flt3-ligand have been identified. The predominant biologically active form is anchored to the cell surface as the extracellular domain of a transmembrane protein (209a.a.). The membrane-bound isoform can be proteolytically cleaved to generate a biologically active soluble isoform.

Biological Activity

The ED₅₀ of ≤ 1.0 ng/ml was determined by the dose dependent stimulation of the proliferation of human AML5 cells. It corresponds to a specific activity of ≥ 1x10⁶ units/mg.

Reconstitution

We recommend a quick spin followed by reconstitution in distilled, sterile water to a concentration of 0.1-1.0mg/ml. This solution can then be diluted into other aqueous buffers and stored at 4°C for one week or -20°C for future use.

Stability

The lyophilized protein is stable for few weeks at room temperature, but best stored at -20°C. Reconstituted Flt3-ligand should be stored in working aliquots at -20°C. Please avoid repeated freeze thaw cycles.
