Description of Human Immunoglobulin G1 (IgG1)

Human Immunoglobulin G1 (IgG1) is a 146 kDa protein. Serum IgG1 constitutes approximately 65% of total immunoglobulin G, making it the predominant subclass in human circulation. This glycoprotein features a 15-amino-acid hinge region that confers intermediate flexibility between IgG subclasses, enabling broad antigen recognition and effector functions. The Fc domain of IgG1 binds high-affinity FcγRI (CD64) via a hydrophobic pocket accommodating Leu235, facilitating immune complex clearance through phagocytosis, antibody-dependent cellular cytotoxicity (ADCC), and complement activation. IgG1’s Fc also interacts with FcγRIII and C1q, making it central to both innate and adaptive immunity. Abnormally elevated IgG1 levels are hallmark in multiple myeloma, where clonal plasma cell proliferation disrupts normal immunoglobulin ratios, often leading to renal complications. Conversely, IgG1 deficiency (<4.9 g/L) predisposes individuals to recurrent bacterial infections due to impaired opsonization of protein antigens like tetanus toxoid7. Autoimmune disorders such as systemic lupus erythematosus (SLE) and Sjögren’s syndrome exhibit IgG1-predominant autoantibodies targeting nuclear antigens, which exacerbate tissue damage via FcγR-mediated neutrophil and macrophage activation. IgG1 also drives type II hypersensitivity reactions, with FcγRIII cross-linking on mast cells contributing to anaphylactic pathways.
