Description of Human Immunoglobulin M (IgM), mu chain

Human Immunoglobulin M (IgM), mu chain is a 75 kDa protein. The IgM heavy chain, known as the mu (µ) chain, is a fundamental component of IgM antibodies and consists of 576 amino acids. Each IgM molecule is composed of five mu chains, forming a pentameric structure that enables robust antigen binding and efficient activation of the complement system. The mu chain is distinguished by its high degree of evolutionary conservation across species, more so than the heavy chains of IgG or IgA, underscoring its essential role in the immune response. Purification of the mu chain is typically achieved by reducing the disulfide bonds that link it to the kappa and lambda light chains, allowing for detailed structural and functional studies. Clinically, abnormalities in the mu chain can be associated with immunodeficiency disorders, such as selective IgM deficiency, and certain B-cell malignancies like Waldenström’s macroglobulinemia. Additionally, the mu chain serves as a target in diagnostic assays for early infection and is a key marker in the study of B-cell development and immune system evolution.
