Description of Human/Murine/Rat Activin A (InCs)

Recombinant human Activin A is a 26.0 kDa disulfide-linked homodimer of two beta A chains, each containing 116 amino acid residues, belonging to the TGF-beta family. It exhibits a wide range of biological activities including regulation of cellular proliferation and differentiation and promotion of neuronal survival. Elevated levels of Activin A in human colorectal tumors and in postmenopausal women have been implicated in colorectal and breast cancers, respectively. The biological activities of Activin A can be neutralized by inhibins and by the diffusible TGF-beta antagonist, follistatin.

 

Biological Activity

The ED₅₀ of ≤ 2ng/ml was determined by its ability to inhibit the proliferation of murine MPC-11 cells. It corresponds to a specific activity of ≥ 5.0 x 10⁵ units/mg.

 

Reconstitution

We recommend a quick spin followed by reconstitution in water to a concentration of 50µg/ml. This will yield a solution of 10mM Citric Acid at pH 3.0 which can be stored 4°C for up tp 4 weeks or -20°C for future use. It can be diluted - preferably into PBS containing 2mg/ml albumin (e.g HSA ultra pure 2835.958.xyz) - and stored at -20°C or -80°C.

 

Stability

The lyophilized protein is stable for a few weeks at room temperature, but best stored at -20°C. Reconstituted Activin A should be stored in working aliquots at -20°C. Please avoid repeated freeze-thaw cycles.
